Analysis of specific diagnostic antigens of Clonorchis sinensis adult worms

Abstract

Aim: To find out the specific antigens for immunodiagnosis of clonorchiasis. Methods: The soluble antigens of Clonorchis sinensis(C.sinensis) adult worms were analyzed by SDS-PAGE and Western blot.Sera for this study included sera from patients with clonorchiasis, paragonimiasis, schistosomiasis, trichinellosis,sera from rats and mice infected with T.spiralis,and sera from normal rats,mice and persons. Results: SDS-PAGE revealed that the soluble antigens of C.sinensis adult worms had 17 protein bands. Western blot results showed that the protein bands with MW 108 000, 71 000, 65 000,63 000,53 000 were cross-reacted with sera from patients with paragonimiasis. All of the protein bands with MW 108 000,94 000,71 000 were cross-reacted with sera from patients with schistosomiasis, rats infected with T.spiralis and patients with trichinellosis. The protein component with MW 56 000 was only reacted with sera from patients with clonorchiasis.Conclusion: The protein component with MW 56 000 is the specific antigen for C.sinensis adult worm.
