Comparison of biological features of CIK cells from healthy and cancer patients

Abstract

Objective To compare the proliferation,phenotype and cytotoxicity of CIK(cytokine-induced killer) cells from two different tissues of healthy subjects and cancer patients.Methods The peripheral blood of healthy subjets and cancer ptients were harvested.The PBMC were isolated by density gradient centrifugation,induced with IFN-γ CD3MAb,IL-1 and IL-2 to prepare CIK cells.The proliferation of the cells were compared by cell counting,the phenotype of CIK cells were analysed by flow cytometry and the cytotoxic activities to K562 cells and LOVO cells were determined by MTT assay.Results The proliferation speed and the cytotoxicity activity of CIK cells originated from peripheral blood of healthy donors was significantly higher than those of malignant tumor patients(P0.01);The percentage of CD3+CD56+of CIK cells originated from peripheral blood healthy donors was significantly higher than those of malignant tumors patients(P0.01).Conclusion The proliferation,percentage of CD3+CD56+ and the cytotoxicity activity of CIK cells originated from peripheral blood of healthy donors is high and it offer experimental evidence for choice of CIK cells originated from differents tissues in immunological treament.
