Abstract

Objective To provide a theoretical basis for the treatment of leukemia.Methods Ficoll-Hypaque centrifugation method was used to harvest mononuclear cells from the peripheral blood of healthy and non-hodykin lymphoma subjects,the umbilical cord blood;then were incubated to cytokine-induced killer(CIK)cells with cytokine.The CIK phenotypes were measured by flow cytometry,the killing efficacy towards K562 cells were detected by MTT assay.Result Mononuclear cells from three different sources were all incubated to CIK cells,the proliferation speed and the cytotoxicity activity of CIK cells originated from umbilical cord blood were significantly higher than those from peripheral blood(P0.01).Conclusion The CIK cells form umbilical cord blood has higher proliferation speed and cytotoxicity activity,and the umbilical cord blood is easy to collect,with low immunogenicity,generous hemopoietic stem cell,less GVHD et al,should give priority to its application in clinical therapy of leukemia.
