Abstract
Objective To study the effect of rhIGF-1 on the proliferation,BMP-2 and Cbfa1 gene expression in rat osteoblast in vitro.Methods Cultured rat osteoblast were treated with rhIGF-1 in different concentrations(0、10、50、100 ng/ml).Cell proliferation was assessed by MTT colorimetric assay,the expression of BMP-2,Cbfa1 gene in rat osteoblast were examined by semiquantitive RT-PCR.Results rhIGF-1 can promote osteoblast proliferation(P0.05) and increase the expression of BMP-2 and Cbfa1 gene in a does-dependent manner(P0.01).Conclusions rhIGF-1 induces the proliferation,differentiation and maturity of osteoblast probably by increasing the expression of BMP-2 and Cbfa1 gene.
