Abstract

Objective:To establish a RP-HPLC method for determination of oxymatrine lipo-some and their entrapment efficiency. Methods: HPLC conditions were as follows:a DimonsilTM ODS column(200mm × 4. 6mm, 5μm), a room column temperature, a mobile phase consisting of 0.1% H3PO4 solution (pH 3.13 adjusted with triethylamine)-acetonitrile (95:5),a flow rate of 1mL·min-1 ,a detection wavelength of 215nm. The free oxymatrine was separated by Sephadex G-50 column from oxymatrine liposome. Results: The linear range for oxymatrine was 20-100μg·mL-1 ( r = 0.9999, M =5),the intra-day and inter-day RSD were2%,the average recovery was 99. 90% -102.0% .Conclusion:A simple,rapid,and accurate method for determination of oxymatrine liposomes and their entrapment efficiency is established.
