Abstract
[Purpose] To explore the relationship between expressions of S-phase kinase-associated protein 2(skp2),cyclin dependent kinase inhibitor protein 27(p27kip1)and gastric carcinogenesis.[Methods] The protein expressions of p27kip1 and skp2 were detected by immunohistochemistry SP method in 69 cases with gastric carcinoma,28 cases with normal gastric mucous membrane were examined as control group.Three cases randomized from 69 cases with gastric carcinoma were examined by reverse transcription polymerase chain reaction method(RT-PCR)for the mRNA expression of skp2 and compared with their normal gastric mucous membranes as control.[Results] The positive expression rate of p27kip1 protein was 46.38% in gastric carcinoma,while that was 96.43% in normal gastric tissue(P0.01).The positive expression rate of p27kip1 protein decreased with lower differentiation,deeper invasion,lymph node metastasis,advanced pathological stage(P0.05).The expression of skp2 mRNA in gastric carcinoma was up-regulated obviously(P0.01).The positive expression rate of skp2 protein was 33.33% in gastric carcinoma,while that was 10.71% in normal gastric tissue(P0.05).The positive expression rate of skp2 increased with lower differentiation(P0.05).There was a negative correlation between the expressions of skp2 and p27kip1 in gastric carcinoma(P0.01).[Conclusions] The examination of p27kip1 and skp2 protein is helpful to clinical diagnosis and prognostic evaluation in gastric cancer.
