Abstract
Objective:To investigate the role of Cks1 protein in gastric tumorigenesis and P27-Kip1 degradation regulated by Skp2. Methods:Flow cytometry was used to detect the expression of Cks1, P27-Kip1,and Skp2 protein in 20 cases of normal gastric tissue and 20 cases of atypical hyperplasia of gastric mucous membrane as well as 54 cases of primary gastric cancers. Results:The expression of Cks1 and Skp2 gradually increased from normal gastric tissue, atypical hyperplasia of gastric mucous membrane to gastric cancers (P0.05). But the expression of P27-Kip1 gradually decreased(P0.05). Expression of Cks1 or Skp2 was negatively correlated with P27-Kip1 (r=-0.649,P0.05;r=-0.732,P0.05,respectively). There was no correlation between Cks1 and Skp2 protein expression (P0.05). The expression of Cks1 in gastric cancer was significantly associated with tumor differentiation (P0.05), but was not associated with the depth of tumor invasion, lymph node metastasis,and clinical stage(P0.05). Conclusion:Cks1 may be involved in human gastric tumorigenesis and ubiquitylated degradation of P27-Kip1regulated by Skp2.
