Abstract

OBJECTIVE:To establish a high-performance liquid chromatography(HPLC) method for determination of plasma concentration of mycophenolic acid in patients undergoing renal transplantation.METHODS:Mycophenolic acid was determined using Diamonsil C18 column(250 mm×4.6 mm,5 μm) with mobile phase consisting of acetonitrile-0.1% trifluoroacetic acid(40∶60) at a flow rate of 1.0 mL·min-1.The column temperature was maintained at 35 ℃ and the UV detection wavelength was set at 215 nm.Plasma samples were treated by precipitation of protein using acetonitrile.RESULTS:The linear concentration of mycophenolic acid was 0.2-50.0 μg·mL-1(r=0.999 6) with minimum detectable concentration of 0.2 μg·mL-1.The average recoveries rates were all greater than 90%,with extraction recovery rates greater than 100%,and relative standard deviation(RSD) of interassay and intrassay all less than 15%.CONCLUSION:The method is proved to be accurate, simple, and recurrent and it is suitable for the blood concentration monitoring and pharmacokinetic study of mycophenolic acid.
