TECHNICS OPTIMIZATION OF TOTAL ALKALOID EXTRACTION FROM PHELLODENDRON AMURENSE

Abstract

Objective: The extraction process of total alkaloid from Phellodendron amurense with different solvents was optimized by the orthogonal design . Methods: P. amurense was designed as material, alcohol and water as solvents, and the total alkaloid from P. amurense was done. The extraction rates of total alkaloid from P. amurense were studied by using single-factor test with different extraction time, extraction temperature, ratio of material to liquid, alcohol concentrations. Based on the results in single-factor test, berberine hydrochloride as index, the optimum processing conditions were given by the orthogonal design the using of L9(33) orthogonal experiment. Results: The optimum processing conditions with water as agent are 100 ℃ extraction temperature, 4 times , 2.5 h extraction time, 1:16 ratio of material to liquid. At these conditions, the content of total alkaloid from Phellodendron amurense is 46.55 mg/g. The processing conditions with ethanol as agent are 70 ℃ extraction temperature, 70% ethanol concentration, 1:12 ratio of material to liquid, 80 min extraction time, 3 times, the content of total alkaloid from P. amurense is 68.49 mg/g at these conditions. Conclusion: The optimized extraction process is rational design, feasible, simple, stable. Alcohol could shorten extraction time, save energy and increase 32.03% extraction rate of total alkaloid than water.
