Effects of Huangqi Shixiao Powder on Caspase-3、Bax and Bcl-2 Expressions in Rats with Acute Myocardial Infarction

Abstract
Objective:To investigate the effects of different dosages of Huangqi Shixiao Powder on protein expression levels of Caspase-3、Bax and Bcl-2 in rats with acute myocardial infarction.Methods:120 male SD rats were randomly divided into 5 groups:model group,sham group,large-dose group(28g·kg-1·d-1,Huangqi Shixiao Powder pretreatment),medium-dose group(14g·kg-1·d-1,Huangqi Shixiao Powder pretreatment),and low-dose group(7g·kg-1·d-1,Huangqi Shixiao Powder pretreatment).After being administered corresponding medicine for 7 days,all groups except sham group underwent coronary ligation of left anterior descending branch to establish acute myocardial infarction models,and sham group only received thoracotomy without coronary ligation.Four and half hours after AMI,myocardium tissues were collected to measure the infarct size by TTC staining and protein expression levels of Caspase-3、Bax and Bcl-2 by western blot.Results:The infarct size and Caspase-3 and Bax expressions decreased significantly in all the treatment groups than that in model group,while Bcl-2 expression increased significantly than those in model group,P0.01.There are statistic differences among different dosage of Huangqi Shixiao Powder groups,P0.05,and the large-dose group receives a better effect than the others in the matter of decreasing infarct size、Caspase-3 and Bax expressions and increasing Bcl-2 expression.Conclusion:Huangqi Shixiao Powder could decrease infarct size and inhibit the myocardial apoptosis by decreasing Caspase-3 and regulating the balance of Bax and Bcl-2 expressions.
