Comparison of results between ECLIA and ELISA for serum HBsAg detec-tion

Abstract

Objective To compare the result of electro chemi luminescence im-munoassay(ECLIA)with enzyme linked immunoabsorption assay(ELISA)for serum HB-sAg detection.Methods One-step ECLIA was combined with one-step and two-step ELISA to make simultaneous detection of quantitative antigen.With one-step and two-step ELISA,we detected190serum samples screened by ECLIA divided them into four models:①concentration of HBsAg was1~20COI;②concentration of HBsAg was21~4000COI;③concentration of HBsAg was higher than4000COI;④HBsAg was negative(it's concentration was lower than1COI),but HBeAg and HBcAb were positive.Results Among190serum samples,the positive ratio of one-step ELISA was84.2%,of two-step ELISA was92.6%and of ECLIA was97.4%.Conclusion One-step ELISA is more significantly addected by hook effect than two-step ELISA and ECLIA,and it's sensitivi-ty is also lower than the latter two.Two-step ELISA can avoid hook effect,but its sen-sitivity is lower than that of ECLIA,and it's operation time was also longer than that of ECLIA.
