Abstract

Objective In order to examine the correlation between the expression of heat shock protein 70(HSP70)with proliferation and apoptosis of brain glioma.Methods Culturing brain glioma cell line cells in vitro,inhibiting the expression of HSP70 with quercetin,utilizing flow cytometry,electronic microscopy and biochemical techniques,analyzing the growth,cell cycle of brain glioma after inhibiting the HSP70 expression were all the techneques applied in this study.Results Quercetin inhibited the growth of brain glioma cells cultured in vitro in a concentration dependent manner.FCM analysis showed increase in cells in the G 2/M phase and decrease in the G 0/G 1 phase of the cell cycle.Subploidy peaks were found after treatment.Electron microscopy provided evidence that apoptosis occurred in the tumors of treatment groups.Such as densation and margination of chromatin,nuclei fragmentation.Conclusion There is a correlation between HSP70 immunohistochemical expression and the degree of malignance of glioma.The expression of HSP70 on human glioma is of some prognostic significance.Quercetin markedly inhibites the growth of glioma cells in a concentration dependent manner.
