Abstract
To construct the tissue engineered skin by the cells that were isolated from the skin of patients and cultured in vitro,the structure and function of this kind of skin were investigated by animal model.The keratinocytes and fibroblasts were isolated from scalp and dermal tissue respectively.After these two kinds of cells had been cultured in vitro for a short time,they were combined in a three dimensional culture system and becoming the tissue engineered skin.Then,this kind of skin was transplanted on the full thickness skin wound on the back of BALB/C nude mouse.The structure and function would be investigated by pathological method after wound healing for two months.This kind of tissueengineered skin could be constructed within 2—3 weeks by culturing the keratinocytes and fibroblasts in vitro.After having been transplanted,the full thickness skin wound was healed within 2 weeks in the both groups.The pathological results showed that the structure of the combined cells skin was better than that of monolayer keratinocytes.It is conclused that kind of tissue engineered skin could be constructed by the cells derived from the skin of patients in a short time.It might be useful for the care of the severe burned patients
