Abstract

Objective :To culture and proliferate keratinocytes in vitro,and to construct tissue-engineered skin with the cultured cells as well as acellular dermal matrix for tissue-engineered skin.Methods Human keratinocytes were isolated from child forskin dersertded after circumcision procedure,the cultured cells in great shape were seeded onto the surface of acellular dermal matrix.When it was cultured absolutely inside liquid medium for about a week,the upside surface then was lifted outside exposed to air.In this way,tissue-engineered skin was expected to be constructed in vitro.The cultured cells and construction of tissue-engineered skin were observed.Results The cultured cells were observed growing and proliferating well in vitro.With the cells cultured on the surface of acellular dermal matrix with cell-free,tissue-engineered skin could be construted in vitro.Conclusion Constuction of tissue-engineered skin with human keratinocytes as well as acellular dermal matrix in vitro is feasible.
