Abstract
Objective According to the principle of increasing the sensitivity and reducing the background, three DNA silver staining methods on Polyacrylamide gels electrophoresis (PAGE) were analyzed and evaluated. Methods Serial diluted pBR 322 MspⅠ DNA molecular weight standard were separated by non-denature vertical PAGE. Two nitrate silver and one dimmine silver staining methods have been used. Sensitivity and background of those methods were evaluated. Results The sensitivity of all the band of pBR 322 MspⅠ Marker developed is coequally 25 ng. For the aspect of at least single band developed, the sensitivity of dimmine silver staining methods (6.25 ng) inferior to the others. Conclusion The methods developed by Carlos J. et al. is the fastest (20 minitus). The background of dimmine silver staining methods is the lowest, and it's results is convenient for preservation. While the methods developed by Brant J. et al. have no superiority to the others in application.
