Abstract
[Objective]To establish the determination method of Astragaloside IV in Astragalus spp. dry root by HPLC-UV. In addition,to compare Astragaloside IV content in three life forms Astragalus spp. [Method]A KromasilC18column(4. 6 mm × 250 mm),a mobile phase consisting of acetonitrile: water = 32∶ 68 solution with flow rate1. 0 ml /min and a UV detector at 203 nm were adopted. [Result]Astragaloside IV has a linear limit of 0. 05- 0. 5 mg /ml(r = 0. 999),a mean recovery of ciprofibrate is 98. 216%(RSD = 2. 85%). The content of Astragaloside IV in wild type was higher than Astragalus penduliflorus subsp. mongholicus and Astragalus membranaceus( Fisch.) Bunge).[Conclusion]The HPLC-UV method is relatively stable and simple. It can be used in determination of Astragaloside IV in Astragalus spp.
