Abstract
Objective To investigate optimal streptozotocin dosing regimen in order to establish long-term stable diabetic mice model.Methods Sixty C57BL/6J mice were randomly divided into a control group and 5 model groups(n = 10).Among them,the streptozotocin dose of groups with multiple low-dose induction program(daily intraperitoneal injection for 5 consecutive days) were 60 mg/kg,70 mg/kg and 80 mg/kg respectively.There were 2 model groups with disposable administration program(intraperitoneal injection once only),whose dose of streptozotocin were 160 mg/kg and 200 mg/kg respectively.The control group injected with the same amount of citric acid salt buffer. Random blood glucose,food intake,body weight,the molded rate and mortality were monitored in mice of each group in the different periods.Results Each model mice showed the typical performance three more and one less .The blood glucose level of model mice was significantly higher than that of the control group.The 80 mg group and the 200 mg group molded rate were 100% and 70% respectively at 1st week,but those molded rates were 30% at the 12th week because those mortality was significantly increased after the 4th week.The blood glucose level of the 160 mg group mice was not enough high,so the molded rate were only 40% and 0 at 1st week and 12th week respectively.The molded rate of 60 mg group was 70% at the 4th week,however,the rate reduced to 30% at the 12th week because of those blood glucose level reduced gradually.Moreover,the blood glucose level fluctuated significantly.The molded rate of 70 mg group was 100% and 70% at the 1st and 12th week respectively,which was significantly higher than that of other model groups.However,the mortality was only 20% at 12th week.Moreover,the blood glucose levels was stable and more than 20 mmol/L.Conclusion Streptozotocin by intraperitoneal injection can well induce elevated the blood glucose level of mice.Multiple low-dose(70 mg/kg) administered program is the optimal program in establishing longterm stable diabetic mice model.
