Abstract
[Objective]To discuss the effect of two doses of alloxan on the preparation of diabetic mice model. [Methods]129 SPF grade male Kunming mice which the body weight ranged from 18 g to 26 g were divided into two dose groups(180 mg/kg and 160 mg/kg).Mice were given alloxan by intraperitoneal injection.The model success rate and death rate of two groups were compared.10 mice from every dose group,10 failure model mice in 160 mg/kg dose group and 10 normal mice were collected,the changes of body weight and blood sugar were observed 7 days after injection.The blood glucose value of 160 mg/kg dose group was tested while feeding to 28 days. [Results]The model success rate and death rate of 180 mg/kg dose group were higher than those of 160 dose mg/kg group,but the difference between two groups was not significant(P0.05).In 180 mg/kg dose group,the blood glucose level of model mice increased rapidly and subsequently declined slowly with weight loss.In 160 mg/kg dose group,the blood glucose level of model mice increased steadily,maintained for a long time,and damages were slight. [Conclusion]After treated with alloxan by intraperitoneal injection,the blood glucose level of 160 mg/kg dose group was stable,and the modeling effect was better than 160 mg/kg dose group.
