Abstract

The cDNA of hTR( human telomerase RNA)gene was cloned by the means of RT-PCR and inserted into the reverse transcription virus vector(PLNCX)to construct the antisense expression plasmid of hTR gene. By using lipofectin-mediated DNA transfection, the ob- rained plasmid WPS successfully expressed in human liver cancer cells (SMMC-7721). All data suggest that telomerase antisense RNA could significantly inhibit the telomerase activity and proliferation of cells,prolong cell's doubling time and induce the apotosis of cells.
