Abstract

[Purpose] To investigate the effect and mechanism of Trametes robiniophila Murr induced apoptosis of human hepatocellular carcinoma cell line MHCC97H. [Methods] MHCC97H cells were incubated with different dose of Trametes robiniophila Murr or 5-Fu. Cell growth and apoptosis were assessed by CCK-8, flow cytometr (FCM) and fluorescence microscopy. The change of Bax, Bcl-2 and p53 expression were detected by cell immunohistochemistry. [Results] The growth of MHCC97H cells was inhibited and the apoptosis was induced by Trametes robiniophila in vitro in a time/dose-dependent manner. Typical morphological changes of early apoptosis could be observed by fluorescence microscopy. Cell immunohistochemistry showed that the expression of Bax and p53 was upregulated and expression of Bcl-2 was downregulated. [Conclusion] Trametes robiniophila Murr can markedly induce apoptosis in human hepatocellular carcinoma cell line MHCC97H. The mechanism might be related to the change of Bax, Bcl-2 and p53 expression.
