AN EXPERIMENTAL STUDY ON INDUCTION OF HUMAN HEPATOCELLULAR CARCINOMA CELL LINE APOPTOSIS BY TRAMETS ROBINIOPHILA IN VITRO

Abstract

Objective To study the effect of Tramets robinophila on the apoptosis of the human hepatocellular carcinoma cells line HepG-2 and provide the experimental basis for Tramets robinophila treating liver cancer in clinic.Methods The effects of Tramets robinophila on proliferation,apoptosis and DNA synthesis of HepG-2 cells were determined by MTT assay,TUNEL,and flow cytometry.Agrose electrophoresis was applied to investigate DNA-LADDER.Results The growth of HepG-2 cells was inhibited by Tramets robinophila in vitro in a time/dose-dependent manner.Tramets robinophila induced apoptosis of HepG-2 cells and reduced their DNA synthesis.Conclusion Extractum trametes robiniophila murr is effective in inhibiting growth of HCC in vitro.The mechanism of antineoplastic effect of trametes robiniophila might be inhibition of DNA synthesis and induction of apoptosis of tumor cells.
