ESTABLISHMENT OF CHEMOSENSITIVITY TESTING OF ORAL CANCER USING THE MTT ASSAY

Abstract

Objective To establish chemosensitivity testing of oral cancer using the MTT assay. Methods MTT assay was performed for chemosensitivity testing of 28 cases of oral cancer and TCa8113, ACC 2 cell lines. The effect of experiment processes on results of trial was analyzed comparatively. Results MTT assay could examine precisely chemosensitivity of oral tumor cell lines and fresh tumor tissue. But these processes of chemosensitivity testing of fresh oral cancer tissue using the MTT assay were noticed specially, such as selecting tumor tissue, preparing tumor cell suspension, culture of tumor cell, incubation time of MTT and centrifuge of formazan. Conclusion Chemosensitivity of fresh oral cancer tissue could be examined precisely using the MTT assay. This chemosensitivity testing using MTT assay had many advantages, such as better repetition, shorter examination periods and choosing of great number of drugs. In this trial processes there was no radioactive pollution.
