Abstract

Objective:To observe the effects of arsenic trioxide(As_2O_3)and cisplatin(DDP) alone or in combination on reverse of resistance in HTB-56/DDP cell lines and apoptosis-inducing effects of arsenic trioxide.Methods:The sensitivity of HTB-56/DDP cells to As_2O_3 and DDP was tested by means of MTT in vitro and the morphological changes of apoptosis were detected by electron microscopy.Results:The resistance of HTB-56/DDP cell line to DDP was 14.65 times higher than that of HTB-56,but the IC50 value of both cell lines to As_2O_3 was similar(P0.05).Cell inhibition by DDP combining with low-dose As_2O_3 was higher than that of administration alone.As_2O_3 could reinforce the sensitivity of HTB-56/DDP cells to DDP and induce apoptosis of cell lines.Conclusion:As_2O_3 might be an effective anti-tumor drug as well as an effective drug for reversal of HTB-56/DDP to DDP.
