Abstract

Objective To establish an HPLC method for the determination of Puerarin in Pueraria lobata of Yuntai Mountain,and provide a scientific base for evaluation of the mild Pueraria lobata in Yuntai mountain.Methods(SHIMADZU)VP-ODS,chromatographic column was 5μm,150 mm×4.6 mm and column temperature was 40℃;mobile phase was alcohol-water(27:73)(containing 0.5% acetic acid);the flow rate was 1.0 ml/min with detective wavelength was 250 nm;the volume of injection was 10μl.Results The linear range of puerarin was 10.0~160.0 μg·ml~(-1),Y = 65.713X-11.932,r =0.999 5,the average recovery was 98.72%,RSD= 1.52%.Conclusion There is no significant difference in the determination of Puerarin between mild Pueraria in Yuntai mountain and commercial Pueraria,and they are similar in inner quality.Wild Pueraria lobata can be used as a substitute for commercial pueraria roots.
