Expression and significance of fragile histidine triad in sporadic colonic cancer

Abstract

Objective To investigate the expression of fragile histidine triad (FHIT) gene protien and it's possible relationship between FHIT expression and clinicopathological indices in sporadic colon cancer. Methods Using anti-human FHIT protein antibody of rabbit and citrate-micro wave-SP immunohistoehemical method, FHIT expressions were detected in the specimens of 74 cases of colon cancer,26 cases of normal tissuses and 26 cases of benign adenoma respectively. It's clinicopathologiecl indices (Dukes stage etc) and five years survival rate were analyzed. Rusults FHIT gene expressions were negative in l case of 26 cases(3.8%) of normal colonic tissues,in 7 of 26 cases(26.9%)of colonic adenoma tissues, in 35 of 74 cases(47.3%) of colon cancer tissues respectively.There were significant difference in the former three kings(P0.05). FHIT protein low expressions were not associated with age,sex,tumor position and size,serum CEA level and histologic type. However, there were significant difference in the degree of differentiation,pathologic stage, lymph node metastasis, distant metastasis and 5 years survival rate(P0.05). Conclusion Reduction of FHIT expressions may play an important role in the development and progression of the colon cancer. And it may become a new prognostic marker in colon cancer.
