Abstract
In this article ,we choose ten acid and bile salt tolerance Lactobacillus acidophilus through intimating state of people's stomach. According to the nutritional needs of Lactobacillus acidophilus basis culture medium and growth promoting factors were optimized by quadratic design. The optimum basis culture medium composition was as following:1.0% peptone, 0.5% glucose, 1.5% soya oligofructose, 1.0% yeast extract. The optimum growth promotingfactors were as followlng: 12% tomato juice. Growth curve with optimized stimulatedculture were determined by following the change of pH and OD. According to the growth curve, the terminal time was at 16 hours. Lactobacillus acidophilus got to 2.2×1012 cfu/mL in this culture for 16hours at 37 ℃.
