Abstract
Objective To observe the effect of transplantation of rat bone marrow stem cells in the treatment of acute injury of liver.Methods Intragastric administration was taken to 50 rats with CCL4-AAF by which the acute hepatic injured models were made.Bone marrow mononuclear cells which were isolated from bone marrow with lymphocyte separating medium were labeled by PKH26.The rats were divided into 2 groups.The rats were autotransplanted with 0.4 mL bone marrow stem cells suspension via portal vein.The levels of serum alanine aminotransferase(ALT,AST),total bilirubin(TB),prothrombin time(PT),and albumin(ALB)were examined before transplantation and 3 days,1,2 and 4 weeks after transplantation.Four weeks after transplantion,the liver tissues were taken from the rats of every group and cut into frozen sections.The immunohistochemistry staining by fluorescein isothiocyanate was done to the sections for detection of the PKH26-labeled cells.Results The indexes of liver function did not show remarkably changes in 3 days,7 days after transplantation(P0.05).ALT,AST and ALB showed remarkably changes in 2 weeks after transplantation(P0.05).Significant improvement in liver function indexes was found after 4 weeks(P0.05).Red fluorescence of PKH26 in the sections was observed.The number of PKH26-labeled cells was(57.2±6.18).There was significant difference between the experiment group and control group(P0.05).Conclusion Under injured liver status,bone marrow stem cells can be migrated to the liver.Transplantatia is a safe and effective way of treatment,and it can significantly improve the indexes of liver function of rats with acute injury of liver.
