Abstract

Objective To document the polymorphism of molecular type and serotype of Salmonella isolated from person in preventive physical examination by creating database aided with PFGE.Methods Twenty-one Salmonella stains were identified and serotyped by diagnostic serum.Then the total genomic DNA of the isolates was digested with restriction enzyme Xba I,and run pulsed-field gel electrophoresis(PFGE).The DNA fingerprints of different stains were analyzed by Bionumerics5.1 software to perform clustering.Results The 21 Salmonella strains belonged to three subgroup of O:4(B),O:9(D1),O:3,10(E1) and 8 serotype;S.Derby,accounted for 33.3%(7/21);S.Agona were 19.1%(4/21) and S.Typhimurium 14.3%(3/21);The DNA fingerprints of 18 Salmonella strains belonged to 15 PFGE pattern combinations.Similarity value of these strains was in the range of 53.9%~100%.One serotype Salmonella showed several PFGE patterns combinations.Conclusion PFGE pattern combinations of this study show polymorphism;The PFGE technology is suitable for molecular typing of Salmonella in preventive physical examination.
