Abstract
Objective The aim of this study was to investigated the effect of Penehyclidine hydrochloride on the expression of ICAM - 1 in lung in vivo after lung ischemia - reperfusion in rats.Methods Thirty - six male Spragne - Dawley rats were randomized to three groups (twelve control,twelve ischemia-repedusion,twelve after Penehyclidine Hydrochloride).I/R injury consist of 45 min of lung cross -clamping followed by 5 hours of reperfusion;after Penehyclidine Hydrochloride preceding I/R;control animals had a thoracotomy only.Lung tissue wet - to - dry weight ratio and myeloperoxidase concentration were markers of edema and neutrophil se- questration,respectively.Superoxide dismutase activity were assessed,also.Expression of ICAM-1 in lung was measured using im- munohistochemistry,observing the changing of lung tissue in pathology under Electron Microscopy and Light Microscopy.Result Ex- pression of ICAM - 1 significantly decreased after Penehyclidine Hydrochloride,the myeloproxidase activity decreased too,and super- oxide dismutase activity increased.Conclusion Penehyclidine Hydrochloride could inhibit the expression of ICAM-1,and reduce neutrophil -infiltration in lung,and attenuate uhrastructure injury of lung tissue,then attenuate pulmonary edema.
