Abstract
Aim To investigate the effects of sodium valproate(VPA) on the proliferation and the apoptosis of acute myeloid leukemia cell line HL-60.Methods After treated with various concentrations of VPA,cell growth inhibition rates were determined by CCK-8 assay;The morphologic changes of HL-60 cells induced by VPA were observed under light microscopy with Wright-Giemsa staining.Cell cycle and cell apoptosis were analyzed by cytometry(FCM).Results The proliferation rate of HL-60 cells in VPA-treated group decreased in a time-concentration dependent manner.After treated with 2.0mmol /L VPA for 48h,condensation of cells and nuclear chromatin,disintegration of nuclear chromatin,and apoptotic bodies were observed under microscope;The apoptosis rate was significantly higher in the cells treated with 1.0 and 2.0mmol /L VPA for 48 h than in untreated cells[(1.25±0.457%),(3.27±0.984%) and(10.3±3.4%),P0.05 ];After treated with VPA,cell cycle was arrested at G0/G1 phase(P0.05).Conclusions VPA could inhibit the proliferation and induce apoptosis of HL-60 cells in a time-dose dependent manner.It may be related to cell cycle arrestting.
