Abstract
OBJECTIVE:To investigate the rule of VPA on the proliferation and apoptosis of human lung cancer cell line-A549.METHODS:Cultured A549 cells were treated by VPA at the concentrations of 0.5,1.0 and 2.0 mmol/L for 48 hours.The morphological and quant changes of A549 cells treated by VPA were observed through a microscope and the proliferation was predicted by methyl thiazolyl tetrazolium(MTT).The expression of Caspase-3 was detected by immunocytochemistry.The change of cell cycle was detected by flowcytometry.RESULTS: The population of the cells decreased and the shape and size changed after being treated with VPA.VPA could inhibitate the growth of A549 significantly,F=28.312,P0.05.The inhabitation rate,apoptosis rate and the percentage of G1 phase in cell cycle of A549 increased significantly,and S phase decreased significantly,P0.05.With the increasing of the concentrations of VPA,the inhabitation rate increased.The expression of Caspase-3 increased while the concentration of VPA increased.CONCLUSION:VPA can significantly supress the growth of A549 and induce apoptosis and cell cycle arrest in G1 phase effectively.
