Abstract
Objective To observe the effect of cryopreservation on platelet aggregative reaction by combination use of 0.02 dimethyl sulfoxide(DMSO) plus a mixture of second-messenger effectors(ThromboSol). Methods Platelet concentrates were treated with 0.02 DMSO, 0.06 DMSO and 0.02 DMSO plus ThromboSol, respectively, and then placed in liquid nitrogen. Having been frozen for one, three and six months, the samples were thawed and analyzed. The platelet counts, ADP and arachidonic acid(AA)-induced aggregative reaction were detected. Results The platelet counts of the 0.02 DMSO-plus-ThromboSol group were similar to those of the 0.06 DMSO group(F=3.67-5.22,q=0.87-1.89,P0.05), while those of the two groups were higher than those of the 0.02 DMSO group (q=3.43-4.26,P0.05). In comparison to the fresh platelets, the number and functional activity of the frozen platelets of these three groups declined significantly (F=3.67-5.22, q=3.16-4.34,P 0.05). Conclusion The second-messenger effectors could reduce the DMSO concentration used in cryopreservation.
