Abstract
Objective To study the recovery of platelet count of platelets following cryopreservation with various doses of second-messenger effectors(thrombosol:amiloride,sodium nitroprusside,adenosine) and 2% DMSO as cryopreserving solution.Methods Part 1,fresh platelet concentrates were divided into 6 groups,using 2% DMSO,6% DMSO,2% DMSO plus thrombosol and 2% DMSO plus one of the contents of thrombosol as cryopreserving solution.Part 2,fresh platelet concentrates were divided into 10 groups,using 2% DMSO,6% DMSO,2% DMSO plus thrombosol and 2% DMSO plus various doses of amiloride and sodium nitroprusside as cryopreserving solution. After frozen for 1 week or 1 month at -80℃, all groups were thawed and tested for platelet count. Results Part 1,there was no difference between adeno-sinegroupand2%DMSOgroup(P0.05),butthesetwogroupsweresignificantlylowerthanthoseofother4groups (P0.05 or P0.01). Part 2,half dose of amiloride and sodium nitroprusside group was significantly higher than that of 2% DMSO group (P0.01),and had no difference with that of 6% DMSO group or thrombosol group(P0.05).Conclusion 2% DMSO plus half dose of amiloride and sodium nitroprusside as cryoprotective agent have the same effect as 6% DMSO or 2% DMSO plus thrombosol on protection against platelet count decrease of thawed platelet.
