Effect of assisting metal and calming wood,nourishing lung yin and tonifying liver and kidney on systolic pressure,ET-1,NO and Ang II in renal hypertension rats of yin deficiency with yang hyperactivity

Abstract

Objective To explore the antihypertensive mechanism on renal hypertension(RH) of yin deficiency with yang hyperactivity in the treatment of nourishing lung yin and tonifying liver and kidney.Methods Seventy-eight healthy male SD rats of 2 month old were screened and randomly divided into a sham operation group(n =13) and an operation group(n =65).RHRs were operated in the operation group and given by Fuzi decoction for 4 weeks after successful operation,and RHRs of yin deficiency with yang hyperactivity were established by regular fight,then randomly divided into a model group(Group M),a captopril group(Group C),a nourishing lung yin group(Group L),a tonifying liver and kidney group(Group K) and a nourishing lung yin and tonifying liver and kidney group(Group A).All the medicine groups were intragastric administrated once at AM 8:00 every day,while the sham operation group and the Group M were treated with equivalent normal saline,for 4 consecutive weeks.Blood pressure(BP) was measured at 24h after the last administration;8 mL of blood was collected from aorta abdominalis for detecting concentration of serum NO,plasma ET-1 and Ang Ⅱ.Results After 4-week treatment,BP in the Group C,K and A was lower than that in the Group M and L,and that in the Group A was lower than that in the Group C,with significant difference(P 0.05).When comparing the Group M with the sham operation group,comparing the Group C,K and A with the Group M,there was significant difference in AngⅡ,ET-1 and serum NO(P 0.05);in the Group L,plasma Ang Ⅱ and ET-1 were decreased,while the serum NO was in increased,with in significant difference(P 0.05);when comparing the Group C,K and A with the Group L,the Group A with the Group C,there was significant difference(P 0.05).Conclusion The antihypertensive mechanism on hypertension of yin deficiency with yang hyperactivity in the treatment of nourishing lung yin and tonifying liver and kidney is decreasing the formation of ET-1 which will enhance the myocardial contractility,increase heart stroke and thicken vessel wall;increasing the production of NO to inhibit the thickening of vessel wall;decreasing the formation of Ang Ⅱ which will lead to vasoconstriction,thickening of vessel wall and reduced ventricular compliance.
