Effect of Granulocyte-macrophage Colony Stimulating Factor on Diabetic Wound Healing

Abstract

[Objective] To explore the effect of granulocyte-macrophage colony stimulating factor on diabetic wound healing.[Methods] Seventy C57BL/6 mice were divided into wild mice group(control group,n=35)and diabetes model group(DM group,n=35).All mice received skin wound(0.8 cm×0.8 cm)in each side of midline after deep anesthesia.The wound sites were digitally photographed,and sampled at the corresponding time stage.Observation of wound tissue healing was conducted,and the rate of wound healing was calculated simultaneously.The content of GM-CSF in the wound sample was measured by ELISA.[Results] Wound healing rate in DM mice was significantly declined compared with that in the control mice group from the 3rd day after wound formation,especially within 7 days.The levels of GM-CSF expression increased obviously in both 2 groups in the 1st day after wound formation.In the control group,the levels of GM-CSF expression were significantly higher than DM group in the 1st day and 3rd day after wound formation.[Conclusion] The low-expression of GM-CSF may be associated with the reduction of inflammatory cells infiltration in early stage of wound healing.
