The Effect of Nuclearl Factor-kappa B on Severe Acute Pancreatitis of Rats

Abstract

Objective To investigate the effect of nuclear factor-kappa B(NF-κB)in pathogenesis of severe acute pancreatitis(SAP)in rats.Methods The modified Schmidt's method was used to reproduce SAP model in rats(inflicted with 5% sodium taurocholate by the method of retrograde pancreatie injection),ninety SD rats were randomly divided into Sham group,SAP group,and Pioglitazone Interference group.Plasma level of amylase and ascites level of amylase were recorded by staining,meanwhile the pathology of pancreatic tissue was graded.Western blot and ELISA were used to detect the expression of NF-κB and the content of IL-6.Results The concentration of serum amylase and ascite amylase,the pancreatic histologic score in SAP group were significantly higher than those in sham group(P0.01);The expression of NF-κB in SAP group was persistencely upgraded after 3 hour and there was significant difference with 3 h and 6 h,while there was low expression in sham group(P0.01).Pioglitazone Interference could significantly inhibit the expression of NF-κB compared to the SAP group(P0.01).The levels of IL-6 increased significantly in SAP group,however,but in Pioglitazone Interference group decreased significantly at the corresponding time.Conclusion NF-κB is involved in the inflammation response of SAP.The expression of NF-κB has a positive correlation with pathology damage of pancreatic tissue.Inhibition of the expression of NF-κB in pancreatic tissue could be helpful for treatment of SAP by downregulating the above cytokines.
