Abstract
Objective To observe the effects and mechanism of aescion on nuclear-kappa B(NF-κB) and serum tumor necrosis factor(TNF-α) expression in severe acute pancreatitis(SAP) in rats and investigate the mechanism that aescion cure SAP.Methods Ninty-six male Wistar rats(250~300 g) were randomly divided into 3 groups:the sham operation group(n=32),the SAP group(n=32) and the aescion therapy group(n=32).Eight rats in each group were killed at the 3rd h,6th h and 12th h after the establishment of the model to detect the activity of NF-κB in pancreas and the expression of serum TNF-α,pathological score of pancreas tissue was calculated at the same time.The other rats in each group were used to observe the mortality in 24 h.Results NF-κB of pancreas in SAP group was increased compared to that in the sham operation group(P0.05),and serum TNF-α increased much more compared to that in SAP group(P0.05).NF-κB expression,serum TNF-α,pathological impairment in pancreas and mortality at 24 h in therapy group all decreased compared to those in SAP group(all P0.05).Conclusion Aescion plays a role in the treatment of pancreatitis.It may depress the expression of NF-κB.Decreasing the over-release of cytokine TNF-α may be one of the mechanism to treat SAP.
