Plant Regeneration Induced from Calli of Explants of Kyoho Grapevine

Abstract

The Kyoho Grapevine was used to study regeneration.Callus were induced from leaves of grapes,differentiated and regenerated into plantlets through organogenesis.The results showed that with 0.1% HgCl2 to disinfect explants for 7min,the rate of pollution and the rate of browning were moderation,and the survival rate was the highest,which was 30.0%.Best medium for the differentiation was MS+6-BA1.0mg/L+NAA0.01mg/L;Leaf callus induction medium of MS additional 5mg/L 6-BA and 0.5 mg/L NAA of leaf callus induction medium was more appropriate;MS+6-BA0.5mg/L+NAA0.05 mg/L was the most suitable for grape adventitious buds callus tissue regeneration;1/2MS+IBA 0.2 mg/L was the ideal rooting medium,and the root length was moderate,the number of root was large.The initial cultivation regeneration system of Kyoho Grapevines was established.
