Abstract

Objective To investigate the protective effect of Curcumin on apoptosis of PC12 cells induced byβamyloid(Aβ)25-35.Methods Differentiated PC12 cells were treated with different concentrations of Aβ25-35 and Curcumin for 24 h.Cell viability was measured by MTT assay and cell apoptosis was detected by Annexin-V staining and flow cytometry.Results The cell viability was significantly decreased 24 h after treatment with Aβ25-35 (P0.01);the decrease was in a dose-dependent manner.Curcumin(5-20μmol/L)showed obvious inhibitory effect on the apoptosis induced by Aβ25-35.Compared with Aβ25-35(20μmol/L)group,cell apoptosis in Curcmin (5μmol/L) plus Aβ25-35 (20μmol/L)group was significantly lower(P0.01).Conclusion Aβ25-35 can induce neuronal apoptosis and Cucumin can antagonize the cell apoptosis induced by Aβ25-35.(Shanghai Med J, 2007,30.843-846)
