Abstract
Objective To investigate the inhibitory effect of matrine on human cervical carcinoma Hela cells in vivo.Methods Cultured Hela cells were treated with different concentrations of matrine solution(0.25~4.0mg/ml)for 24~72 h.MTT assay was used to evaluate the cytotoxic effect of matrine on Hela cells.Flow cytometry was used to detect the apoptosis of Hela cells.Cell morphology was observed with transmission electron microscope.Results Different concentrations of matrine were cytotoxic to Hela cell in a time-dependent and dose-dependent manner.The FCM analysis showed that the apoptosis rate of Hela cells was increased after treatment of matrine.Hela cells presented the characteristic morphological changes of apoptosis under inverted microscope and transmission electron microscope.Conclusion Matrine can inhibit the proliferation of human cervical carcinoma Hela cells in vitro,which is associated with cell apoptosis.
