A experimental study on the optimal dose of bone marrow in mice undergoing allogeneic bone marrow transplantation

Abstract

Objective To study the optimal dose of bone marrow in mice undergoing allogeneic bone marrow transplantation(BMT).Methods After treated with total body irradiation(TBI) with X-ray 7.0 Gy,the female BALB/c mice aged 6 to 8 weeks old were divided into groups of A,B,C and D according to different doses of bone marrow from male C57BL/6J mice aged 6 to 8 weeks old(1×106 ,5×106 ,1×107 and 2×107 ),which were infused within 4 hours.Survival rates,change of peripheral blood leukocytes of all groups were observed regularly.H-2Kb cells,T and B leukomonocytes were detected by flow cytometry.Results The survival rate of mice in group C was lower than that in the other groups(P 0.05).Chimerism rates in groups of A and D were decreasing after chimerism rate of 95% reached.The levels of hematopoietic and immune recovery in group D recovered to the normal level earlier than those in the other groups.Conclusion Within certain range of cell numbers,the more the bone marrow cells are infused,the more are the advantages for reconstitution of hematopoiesis and immunity.On the basis of evaluating survival rates and level of engraftment,a dose of 5×106 bone marrow cells may be taken as an apropriate amount for allogeneic BMT.
