Abstract
Objective To explore the effect of nonmyeloablative conditioning regiment in graft-versus-host disease ( GVHD) after bone marrow transplantation( BMT) in rat models. Methods SD rats were transplanted with Wistar rats in the allogeneic bone marrow transplantation model. SD rats were divided into 2 groups: group A were conditioned with fludarabine for 4 d at a dose of 1 mg/kg on days 4 , 3 , 2 , 1 prior to transplantation, and received total body irradiation (TBI) 2 Gy 4 h prior to BMT and group B were exposed to a dose of 6 Gy TBI on the same time. All the recipient rats of the test group were injected with donor bone marrow cells at a dose of 3. 8 × 10 bone marrow cells per mouse through the tail vein. Rats in the control group were injected with the same volume of saline. Afterwards we observed the GVHD of both groups. Results The survival time of rats which received fludarabine and 2 Gy TBI were longer, and the severity of GVHD was lower. Conclusion Nonmyeloablative allogeneic bone marrow transplantation could effectively prevent GVHD. This conditioning regimen could broaden the application of allogeneic bone marrow transplantation.
