Abstract

【Objective】To investigate the effect of Rapamycin, a inhibitor of the mTOR, on the viability of the colon carcinoma cell line Lovo. 【Methods】The cultured Lovo cells were treated with different concentrations of Ra- pamycin (0.5, 1.0, 2.5 and 5.0 μg/ml) for different time. The proliferation of Lovo cells was detected by MTT method. Cell apoptosis was detected by flow cytometry. The levels of Bcl-2 protein were determined by Western blot. 【Results】Rapamycin inhibited the proliferation of Lovo cells in a concentration- and timedependent manner. Rapamycin induced the apoptosis of Lovo cells in a concentration-dependent manner. The levels of Bcl-2 protein were reduced gradually with the increase of concentration or action time. 【Conclusion】Rapamycin can induce the apoptosis of the colon carcinoma cell line Lovo, reduce the expression of Bcl-2, and inhibite the proliferation of colon carcinoma.
