Expression of p-BAD and p-AKT in Cervical Carcinoma Carcinogenesis Progression and Its Significance

Abstract

Objective To study the expression and significance of p-BAD and p-AKT in cervical carcinoma progression,namely in normal cervical tissue,cervical epithelial dysplasia(CINⅠ,Ⅱand Ⅲ)and cervical carcinoma. Methods The expression of the two proteins was examined in 100 paraffin-embedded cervical tissue specimens by using immunohistochemcal staining and 30 freshly-taken cervical carcinoma and adjacent cervical tissue specimens by Western Blotting. The relationship between the expression of the two proteins and clinicopathological factors was analyzed. Results The positive rates of the p-BAD expression and the p-AKT expression showed a gradual increase in cervical cancer progression. The expression of p-BAD and p-AKT proteins showed a significant difference at CIN Ⅲ level and also showed a significant difference between carcinoma in situ group and cervical carcinoma group(P=0.036,0.048). No obvious correlation was found between the expression of the two protein and the age of the patients. However,the expression of the two protein had a correlation with clinical stage and histological grade(P=0.037,0.018;0.001,0.002,respectively),and the lower the histological grade was,the higher the positive rate of their expression was. The p-AKT protein expression was related to pelvic lymph node metastasis(P=0.023). Western Blotting analysis showed that the levels of of the two proteins in cervical cancer were higher than those in the paraneoplastic tissue(P=0.002,0.003). Conclusion The levels of the two proteins could suggest,to some extent,the potential transformation of cervical intraepithelial neoplasia to cervical cancer.
