Effect of Resveratrol on Growth and Apoptosis of Human Esophageal Cancer Cells

Abstract

Objective: To investigate the effects of reservatrol on growth and apoptosis of human esophageal carcinoma cells.Methods: Human esophageal carcinoma cells(EC9706) were cultured in DMEM culture medium.The cells were treated with reservatrol at 0.2,0.4,0.8,1.6 and 3.2 mmol/L for 24,48 and 72 h in complete cells medium.Proliferation,apoptosis and the cell cycle distribution of EC9706 were assessed by MTT assay and flow cytometry.Induction of apoptosis was further detected using transmission electron microscope.Results: Resveratrol 3.mmol/L inhibited the growth of EC9706 cell line in a dose-and time-dependent manner.The inhibition rate of proliferation was 86.73%,and the rate of apoptosis was 47.53%.After the treatment for 24,48 and 72 h,50% cytotoxin concentration and the maximal non-cytotoxin concentration was(0.32,) 0.08 and 0.09 mmol/L,respectively.Resveratrol in 0.2-3.2 mmol/L also increased the ratio of G_(0)/G_(1),decreased the proportion of cell in S phase and induced apoptosis.Observation under the transmission electron microscope revealed notable apoptosis in the EC9706 cell line.(Conclusion:) Resveratrol can inhibit the growth of EC9706 and induce the apoptosis and G_(0)/G_(1) arrest.Resveratrol may provide a new possible treatment method for esophageal cancer.
