Inhibition of resveratrol on the proliferation of melanoma cells

Abstract

OBJECTIVE: To explore the influence of resveratrol on the growth,cell cycle and apoptosis of melanoma cell lines WM239.METHODS:The effects of resveratrol on the proliferation of WM239 cells were analyzed with MTT assay.The cell cycle and apoptosis was analyzed using flow cytometry with Annexin-V and PI staining.RESULTS: The proliferation of WM239 was inhibited when the cell line was exposed to 12.5-200 μmol/L resveratrol for 48 h and 72 h.The inhibition rate was higher than 50% when the cell line was exposed to above 50 μmol/L resveratrol for 72 h.The inhibition was positively correlated to drug concentration.Exposed to 0,25,50 and 100 μmol/L resveratrol for 48 h,the apoptosis rate were 0.1%,9.7%,8.9% and 19.3%.G0/G1 phase cells were 71.9%,77.3%,81.0% and 83.7% respectively.S phase cells were 21.5%,17.9%,16.0% and 14.6%.G2/M phase cells were 6.5%,4.8%,2.9% and 1.8%.The cell cycle was blocked in G1 phase.The cells of S and G2 phase decreased in a dose-dependent manner.CONCLUSION:Resveratrol could inhibit the proliferation of WM239 cells,affect the cell cycle and cause the apoptosis of WM239 cells.
