Effects of PDGF-BB and bFGF on ALP activity in human periodontal ligament cells:an in vitro study

Abstract

Objective To explore the effects of platelet derived growth factor-BB(PDGF-BB)and basic fibroblast growth factor(bFGF)on alkaline phosphatase(ALP)activity in human periodontal ligament cells(hPDLCs)in vitro.Methods The third generation hPDLCs were cultured in vitro,and then the cells of the fourth to eighth generation were divided into control group and experimental groups.The hPDLCs were incubated with pure cell culture fluid in control group,but with PDGF-BB of 0.1,1,10,100 ng/ml,bFGF of 1,10,50,100 ng/ml,100 ng/ml PDGF-BB+50 ng/ml bFGF,0.1 ng/ml PDGF-BB+1 ng/ml bFGF in experimental groups,respectively.After culture for 3 d,half of hPDLCs was splitted.All groups were divided into lysate subgroup and culture subgroup.ALP activity was detected by enzyme dynamics method.Results At 3 d,OD value of PDGF-BB in lysate group and in culture group peaked at the concentration of 0.1 ng/ml and 1 ng/ml,respectively.OD value of bFGF in lysate group and in culture group peaked at the concentration of 1 ng/ml and 10 ng/ml,respectively.The combination of two factors showed better effect on enzyme activity than in control group.Conclusion Both PDGF-BB and bFGF could enhance the ALP activity significantly.
