Abstract
Objective:To observe the effect of(Saliva, Bupleuri, sparganii, giant knotweed rhizome, nutgrass galingale rhizome)constitute ChaiHu ShuGan decoction on tumor necrosis factor-α(TNF-α) and interferon-γ(INF-γ) in serum of rats with hepatic fibrosis. Methods: The eighty rats were divided into eight groups randomly,every group included ten rats. They were control group, model group, traditional Chinese Drugs JinSanE preventive group, ChaiHu ShuGan decoction preventive group, traditional Chinese Drugs JinSanE therapeutic group, ChaiHu ShuGan decoction low dosage, middle dosage, high dosage therapeutic group. Except control group, the other groups were given 40% carbon tetrachloride liquid paraffin to make model of hepatic fibrosis .Traditional Chinese drugs JinSanE preventive group and ChaiHu ShuGan decoction preventive group were administrated traditional Chinese drugs JinSanE and ChaiHu ShuGan decoction 0.6 g/100 g and 1.0 g/100 g respectively, when the model began to make. Four weeks later, the traditional Chinese Drugs JinSanE therapeutic groups, ChaiHu ShuGan decoction low dosage, middle dosage, high dosage therapeutic group were administrated traditional Chinese Drugs JinSanE 0.6 g/100 g, ChaiHu ShuGan decoction 0.5, 1.0, 2.0 g/100 g respectively eight weeks. The concentration of TNF-αand INF-γin the serum of rats in every group were detected by enzyme linked immunosorbent assay (ELISA). Results: Compared with control group, the concentration of TNF-αin serum of model group increased obviously(P0.01). Compared with model group the concentration of TNF-αin serum of JinSanE preventive group, therapeutic group decreased obviously(P0.05~0.01). Compared with model group, the concentration of TNF-αin serum of ChaiHu ShuGan decoction preventive group decreased obviously(P0.05). Compared with model group, the concentration of TNF-αin serum of ChaiHu ShuGan decoction low dosage, middle dosage, high dosage therapeutic group decreased obviously(P0.05~0.01). Compared with ChaiHu ShuGan decoction low dosage therapeutic group and ChaiHu ShuGan decoction preventive group, the concentration of TNF-αin serum of ChaiHu ShuGan decoction high dosage therapeutic group decreased obviously(P0.05). Compared with control group, the concentration of IFN-γin serum of model group increased obviously (P0.01). Compared with model group ,the concentration of IFN-γ in serum of ChaiHu ShuGan decoction middle and high dosage therapeutic groups decreased obviously (P0.01). Compared with ChaiHu ShuGan decoction low dosage therapeutic group, preventive group, the concentration of IFN-γin serum of ChaiHu ShuGan decoction high dosage therapeutic group decreased obviously(P0.05). Conclusion: ChaiHu ShuGan decotion could decrease the concentration of TNF-α and INF-γin serum obviously. Could it improve hepatic fibrosis or not by decreasing these two cytokines needs further research.
