Abstract
[Objective] The aim was to screen and optimize the liquid fermentation medium and culture condition of Coprinus comatua.[Method] With C.comatua as tested material,extracellular polysaccharide and mycelial biomass as indexes,the liquid fermentation medium formula and culture condition of C.comatua were screened and optimized by single factor experiment and orthogonal test.[Result] The optimal composition of mediums for C.comatua liquid fermentation producing extracellular polysaccharide were as follows:2.0% glucose,0.75 % yeast extract and 0.2% CaCl2,The optimal composition of fermentation mediums for C.comatua liquid fermentation producing mycelia were as follows:2.0% glucose,0.75% yeast extract and 0.2% KH2PO4.The optimal fermentation conditions of C.comatua liquid fermentation producing extracellular polysaccharide and mycelia were as follows: initial pH value of 7.0,inoculation amount of 20.0 %,liquid volume of 120 ml and culture time of 6 d.[Conclusion] The research provided the scientific basis for further development of C.comatua.
