Abstract
The optimal conditions of producing Agaricus blazei mycelia through liquid state fermentation and the process of extracting polysaccharide from the mycelia were determined by pilot tests.Maize meal and sucrose were used as carbon sources,and soybean meal and yeast powder as nitrogen sources in fermentation,the mycelia yield was averagely 16.42%,and the polysaccharide content of mycelia was 25.22%under the conditions:inoculum concentration at 5%,ventilation ratio at 1:0.5(v/v),stirring speed at 150 r/min,and fermentation at 25 ℃ for 72 h.The process of producing polysaccharide of fermentation mycelia was as follows:the mycelia were added with ten fold distilled water,the extraction was made at 100 ℃ firstly for 2 h and then for 1 h,the mixed filtrate was concentrated by 10-kD hollow fiber membrane ultrafiltration concentration ratio of liquid to mycelia reaching 1:1),the concentrated filtrate was added with 2%maltodextrin.and after spray drying the crude polysaccharide yield was 12.64%and its polysaccharide content was 33.47%.
